Sermorelin (10MG) is a synthetic 29-amino acid peptide representing the biologically active N-terminal fragment of endogenous growth hormone-releasing hormone (GHRH 1-44), designated GRF(1-29). Research has established that this truncated fragment retains full biological activity at the GHRH receptor, making it the minimal active sequence for GHRH-R binding and a valuable pharmacological tool for structure-activity relationship studies. Sermorelin binds with high affinity to GHRH receptors (KD 0.2–0.8 nM) on pituitary somatotroph cells, initiating Gαs protein coupling, adenylyl cyclase activation, and subsequent intracellular cAMP elevation — driving GH gene transcription and secretory vesicle mobilization. A key distinguishing feature for research applications is its capacity to maintain physiologically pulsatile GH secretion dynamics subject to normal somatostatin-mediated negative feedback, in contrast to continuous non-regulated GH stimulation models. Its plasma half-life of under 10–20 minutes further supports timed, pulsatile GH secretion modeling in vitro.
