Melanotan 2 (10MG) is a synthetic cyclic lactam analogue of alpha-melanocyte-stimulating hormone (α-MSH) with broad binding activity across MC1R, MC3R, MC4R, and MC5R subtypes — notably excluding MC2R. Key structural modifications relative to endogenous α-MSH include norleucine substitution at position 4 for enhanced metabolic stability, D-phenylalanine substitution at position 7 for increased receptor binding affinity, and cyclization via a lactam bridge between Asp-5 and Lys-10 conferring resistance to enzymatic degradation.
