MOTS-C (10MG) is a 16-amino acid peptide encoded by the short open reading frame of the mitochondrial 12S rRNA gene — making it one of the first peptides known to originate from mitochondrial rather than nuclear DNA. In response to stress or exercise, MOTS-C translocates to the nucleus where it regulates the expression of stress adaptation-related genes with antioxidant response elements (ARE). Its primary signaling mechanism operates through the Folate-AICAR-AMPK pathway: MOTS-C regulates the folate cycle and de novo purine biosynthesis, leading to an increase in AICAR, which phosphorylates and activates AMPK — the master regulator of cellular energy homeostasis. Critically, this allows AMPK engagement without requiring cellular energy depletion, distinguishing it from conventional AMPK activators. This makes MOTS-C a valuable research tool for investigating cellular energy sensing, mitochondrial-nuclear crosstalk, insulin sensitivity signaling, and metabolic adaptation pathways in controlled in vitro systems. Research has further implicated MOTS-C in energy metabolism, insulin resistance, inflammatory response, aging, and aging-related pathologies.
